SPERM MEMBRANE INTEGRITY AND MOTILITY SEXING USE GRADIENT PERCOLL CENTRIFUGATION DENSITY OF BULL USE DIFFERENT CENTRIFUGATION SPEED AND SEMEN VOLUME

ABSTRACT
The research with aim to study sperm membrane integrity and motility during freezing use gradient percoll centrifugation density was carry out. It was expected give information about the best of centrifugation speed and semen volume during freezing.
Research was conducted by experiment method, with Frisien Holstein (FH) bull semen sample. Sexing fresh semen with two centrifugation speed (2250 and 2500 rpm) with semen volume 1, 2 and 3 ml as treatment with 5 replications. Motility and integrity of sperm evaluated before and after frozen. The obtained data analyzed with analysis of variances and Chi-square test.
It was concluded that sperm membrane integrity and motility influenced by centrifugation speed during freezing while semen volume not significant different. The best speed centrifugation on sperm integrity membrane and motility during freezing semen is 2250 rpm for maintain motility and membrane integrity sperm during freezing.
